    Spared Discrimination and Impaired Reversal Eyeblink Conditioning in Patients with Temporal Lobe Amnesia

    The effect of medial temporal lobe damage on a two tone delay discrimination and reversal paradigm was examined in human classical eyeblink conditioning. Eight medial temporal lobe amnesics and their demographically matched controls were compared. Amnesics were able to distinguish between two tones during the initial discrimination phase of the experiment almost as well as control participants. Amnesic patients were not able to reverse the previously acquired two tone discrimination. In contrast, the control participants showed improved discrimination performance after the reversal of the tones. These findings support the hypothesis that the hippocampus, and associated temporal lobe regions, play a role in eyeblink conditioning that becomes essential in more complex versions of the task, such as the reversal of an acquired two tone discrimination.

    Brain Bases of Reading Fluency in Typical Reading and Impaired Fluency in Dyslexia

    Although the neural systems supporting single word reading are well studied, there are limited direct comparisons between typical and dyslexic readers of the neural correlates of reading fluency. Reading fluency deficits are a persistent behavioral marker of dyslexia into adulthood. The current study identified the neural correlates of fluent reading in typical and dyslexic adult readers, using sentences presented in a word-by-word format in which single words were presented sequentially at fixed rates. Sentences were presented at slow, medium, and fast rates, and participants were asked to decide whether each sentence did or did not make sense semantically. As presentation rates increased, participants became less accurate and slower at making judgments, with comprehension accuracy decreasing disproportionately for dyslexic readers. In-scanner performance on the sentence task correlated significantly with standardized clinical measures of both reading fluency and phonological awareness. Both typical readers and readers with dyslexia exhibited widespread, bilateral increases in activation that corresponded to increases in presentation rate. Typical readers exhibited significantly larger gains in activation as a function of faster presentation rates than readers with dyslexia in several areas, including left prefrontal and left superior temporal regions associated with semantic retrieval and semantic and phonological representations. Group differences were more extensive when behavioral differences between conditions were equated across groups. These findings suggest a brain basis for impaired reading fluency in dyslexia, specifically a failure of brain regions involved in semantic retrieval and semantic and phonological representations to become fully engaged for comprehension at rapid reading rates.

    Hyperactivity and hyperconnectivity of the default network in schizophrenia and in first-degree relatives of persons with schizophrenia

    We examined the status of the neural network mediating the default mode of brain function, which typically exhibits greater activation during rest than during task, in patients in the early phase of schizophrenia and in young first-degree relatives of persons with schizophrenia. During functional MRI, patients, relatives, and controls alternated between rest and performance of working memory (WM) tasks. As expected, controls exhibited task-related suppression of activation in the default network, including medial prefrontal cortex (MPFC) and posterior cingulate cortex/precuneus. Patients and relatives exhibited significantly reduced task-related suppression in MPFC, and these reductions remained after controlling for performance. Increased task-related MPFC suppression correlated with better WM performance in patients and relatives and with less psychopathology in all 3 groups. For WM task performance, patients and relatives had greater activation in right dorsolateral prefrontal cortex (DLPFC) than controls. During rest and task, patients and relatives exhibited abnormally high functional connectivity within the default network. The magnitudes of default network connectivity during rest and task correlated with psychopathology in the patients. Further, during both rest and task, patients exhibited reduced anticorrelations between MPFC and DLPFC, a region that was hyperactivated by patients and relatives during WM performance. Among patients, the magnitude of MPFC task suppression negatively correlated with default connectivity, suggesting an association between the hyperactivation and hyperconnectivity in schizophrenia. Hyperactivation (reduced task-related suppression) of default regions and hyperconnectivity of the default network may contribute to disturbances of thought in schizophrenia and risk for the illness.

    Assessing the influence of scanner background noise on auditory processing. I. An fMRI study comparing three experimental designs with varying degrees of scanner noise

    We compared two experimental designs aimed at minimizing the influence of scanner background noise (SBN) on functional MRI (fMRI) of auditory processes with one conventional fMRI design. Ten subjects listened to a series of four one-syllable words and had to decide whether two of the words were identical. This was contrasted with a no-stimulus control condition. All three experimental designs had a duration of approximately 17 min: 1) a behavior interleaved gradients (BIG; Eden et al. [1999] J Magn Reson Imaging 41:13-20) design (repetition time, TR, = 6 s), where stimuli were presented during the SBN-free periods between clustered volume acquisitions (CVA); 2) a sparse temporal sampling technique (STsamp; e.g., Gaab et al., [2003] Neuroimage 19:1417-1426) acquiring only one set of slices following each of the stimulations with a 16-s TR and jittered delay times between stimulus offset and image acquisition; and 3) an event-related design with continuous scanning (ERcont) using the stimulation design of STsamp but with a 2-s TR. The results demonstrated increased signal within Heschl's gyrus for the STsamp and BIG-CVA design in comparison to ERcont as well as differences in the overall functional anatomy among the designs. The possibility to obtain a time course of activation as well as the full recovery of the stimulus- and SBN-induced hemodynamic response function signal and lack of signal suppression from SBN during the STsamp design makes this technique a powerful approach for conducting auditory experiments using fMRI. Practical strengths and limitations of the three auditory acquisition paradigms are discussed.

    Assessing the influence of scanner background noise on auditory processing. II. An fMRI study comparing auditory processing in the absence and presence of recorded scanner noise using a sparse design

    Several studies reported decreased signal intensities within auditory areas for experimental designs employing continuous scanner background noise (SBN) in comparison to designs with less or no SBN. This study examined the source for this SBN-induced masking effect of the blood oxygenation level-dependent (BOLD) response by directly comparing two experimental sessions with the same auditory stimulation, which was presented either with or without recorded scanner background noise (RecSBN). Ten subjects listened to a series of four one-syllable words and had to decide whether two of the words were identical. The words were either presented with a silent background or with added RecSBN. This was then contrasted with either silence or RecSBN. A sparse temporal sampling method was used in both sessions, which enabled us to directly assess the influence of RecSBN without varying scanning parameters, acquisition quantities, or auditory stimulations. Our results suggest that previously reported SBN-induced masking of the BOLD response in experimental designs with SBN might be caused by an interaction between increased baseline levels and nonlinearity effects within auditory cortices. Adding SBN to an experimental condition does not enhance signal intensities to the same degree that SBN does when presented with a silent background, and therefore contrasting an experimental and baseline condition that both have SBN may lead to signal decreases. In addition, our study shows this effect is greatest in Heschl's gyrus, but can also be observed in higher-order auditory areas.

    Resting in peace or noise: Scanner background noise suppresses default-mode network

    Studies have identified specific brain regions that increase activation during rest relative to attention-demanding tasks; these regions subserve the "default mode of brain function". Most of these studies have been conducted in the presence of scanner background noise (SBN). This noise has been shown to lead to altered attentional demands, and thus may modulate the default-mode network. Twelve subjects were examined during a rest condition that was contrasted with an auditory task. Words were presented either with SBN employing a conventional acquisition or without SBN using a sparse sampling approach. The number of experimental and resting trials was equated between the designs. Selecting the images in the condition with SBN that corresponded in time with the images in the condition without SBN made a direct comparison of the default-mode network (rest contrasted with active task) possible. There was typical activation of the default-mode network during rest versus task for both designs. However, SBN suppressed major components of the default-mode network, including medial prefrontal cortex, posterior cingulate, and precuneus. Our results suggest that the default mode of brain function differs when assessed in the presence compared to the absence of scanner noise, with the presence of scanner noise perhaps adding attentional demands that diminish activation changes between rest and task in a nonlinear way within the default network. Further studies are needed to clarify whether the use of a sparse sampling technique might enhance clinical utilities that have been proposed for analysis of the default-mode network.

    Evaluating the validity of volume-based and surface-based brain image registration for developmental cognitive neuroscience studies in children 4 to 11 years of age

    Understanding the neurophysiology of human cognitive development relies on methods that enable accurate comparison of structural and functional neuroimaging data across brains from people of different ages. A fundamental question is whether the substantial brain growth and related changes in brain morphology that occur in early childhood permit valid comparisons of brain structure and function across ages. Here we investigated whether valid comparisons can be made in children from ages 4 to 11, and whether there are differences in the use of volume-based versus surface-based registration approaches for aligning structural landmarks across these ages. Regions corresponding to the calcarine sulcus, central sulcus, and Sylvian fissure in both the hemispheres were manually labeled on T1-weighted structural magnetic resonance images from 31 children ranging in age from 4.2 to 11.2 years old. Quantitative measures of shape similarity and volumetric-overlap of these manually labeled regions were calculated when brains were aligned using a 12-parameter affine transform, SPM's nonlinear normalization, a diffeomorphic registration (ANTS), and FreeSurfer's surface-based registration. Registration error for normalization into a common reference framework across participants in this age range was lower than commonly used functional imaging resolutions. Surface-based registration provided significantly better alignment of cortical landmarks than volume-based registration. In addition, registering children's brains to a common space does not result in an age-associated bias between older and younger children, making it feasible to accurately compare structural properties and patterns of brain activation in children from ages 4 to 11.

    Tracking the Roots of Reading Ability: White Matter Volume and Integrity Correlate with Phonological Awareness in Prereading and Early-Reading Kindergarten Children

    Developmental dyslexia, an unexplained difficulty in learning to read, has been associated with alterations in white matter organization as measured by diffusion-weighted imaging. It is unknown, however, whether these differences in structural connectivity are related to the cause of dyslexia or if they are consequences of reading difficulty (e.g., less reading experience or compensatory brain organization). Here, in 40 kindergartners who had received little or no reading instruction, we examined the relation between behavioral predictors of dyslexia and white matter organization in left arcuate fasciculus, inferior longitudinal fasciculus, and the parietal portion of the superior longitudinal fasciculus using probabilistic tractography. Higher composite phonological awareness scores were significantly and positively correlated with the volume of the arcuate fasciculus, but not with other tracts. Two other behavioral predictors of dyslexia, rapid naming and letter knowledge, did not correlate with volumes or diffusion values in these tracts. The volume and fractional anisotropy of the left arcuate showed a particularly strong positive correlation with a phoneme blending test. Whole-brain regressions of behavioral scores with diffusion measures confirmed the unique relation between phonological awareness and the left arcuate. These findings indicate that the left arcuate fasciculus, which connects anterior and posterior language regions of the human brain and which has been previously associated with reading ability in older individuals, is already smaller and has less integrity in kindergartners who are at risk for dyslexia because of poor phonological awareness. These findings suggest a structural basis of behavioral risk for dyslexia that predates reading instruction.

    Failure of Working Memory Training to Enhance Cognition or Intelligence

    Fluid intelligence is important for successful functioning in the modern world, but much evidence suggests that fluid intelligence is largely immutable after childhood. Recently, however, researchers have reported gains in fluid intelligence after multiple sessions of adaptive working memory training in adults. The current study attempted to replicate and expand those results by administering a broad assessment of cognitive abilities and personality traits to young adults who underwent 20 sessions of an adaptive dual n-back working memory training program and comparing their post-training performance on those tests to a matched set of young adults who underwent 20 sessions of an adaptive attentional tracking program. Pre- and post-training measurements of fluid intelligence, standardized intelligence tests, speed of processing, reading skills, and other tests of working memory were assessed. Both training groups exhibited substantial and specific improvements on the trained tasks that persisted for at least 6 months post-training, but no transfer of improvement was observed to any of the non-trained measurements when compared to a third untrained group serving as a passive control. These findings fail to support the idea that adaptive working memory training in healthy young adults enhances working memory capacity in non-trained tasks, fluid intelligence, or other measures of cognitive abilities.

    A large-scale investigation of white matter microstructural associations with reading ability

    Reading involves the functioning of a widely distributed brain network, and white matter tracts are responsible for transmitting information between constituent network nodes. Several studies have analyzed fiber bundle microstructural properties to shed insights into the neural basis of reading abilities and disabilities. Findings have been inconsistent, potentially due to small sample sizes and varying methodology. To address this, we analyzed a large data set of 686 children ages 5–18 using state-of-the-art neuroimaging acquisitions and processing techniques. We searched for associations between fractional anisotropy (FA) and single-word and single-nonword reading skills in children with diverse reading abilities across multiple tracts previously thought to contribute to reading. We also looked for group differences in tract FA between typically reading children and children with reading disabilities. FA of the white matter increased with age across all participants. There were no significant correlations between overall reading abilities and tract FAs across all children, and no significant group differences in tract FA between children with and without reading disabilities. There were associations between FA and nonword reading ability in older children (ages 9 and above). Higher FA in the right superior longitudinal fasciculus (SLF) and left inferior cerebellar peduncle (ICP) correlated with better nonword reading skills. These results suggest that letter-sound correspondence skills, as measured by nonword reading, are associated with greater white matter coherence among older children in these two tracts, as indexed by higher FA.
